چکیده

Background: Malaria parasites detoxify copious amounts of heme during human infection but enzyme involvement remains unclear. Results: Parasite-infected and uninfected erythrocytes contain indistinguishable low levels of heme catabolites, and a heme oxygenase-like parasite protein binds but does not degrade heme. Conclusion: P. falciparum parasites do not enzymatically degrade heme. Significance: This study comprehensively answers a long-standing question regarding heme metabolism in this important parasite.

Direct Tests of Enzymatic Heme Degradation by the Malaria Parasite Plasmodium falciparum*

Malaria parasites generate vast quantities of heme during blood stage infection via hemoglobin digestion and limited de novo biosynthesis, but it remains unclear if parasites metabolize heme for utilization or disposal. Recent in vitro experiments with a heme oxygenase (HO)-like protein from Plasmodium falciparum suggested that parasites may enzymatically degrade some heme to the canonical HO p...

Cysteine proteinase inhibitors block early steps in hemoglobin degradation by cultured malaria parasites.

Erythrocytic malaria parasites degrade hemoglobin as a source of amino acids for parasite protein synthesis. Cysteine proteinase inhibitors have been shown to block the hydrolysis of globin by cultured parasites, indicating that a malarial cysteine proteinase is required for this process. In the present study, we have evaluated the role of parasite proteinases in earlier steps of hemoglobin deg...

Clinical Pharmacology of the Antimalarial Chloroquine in Children and Their Mothers

Plasmodium falciparum, Plasmodium vivax, Plasmodium ovale, Plasmodium malariae, and Plasmodium knowlesi are the parasites that infect humans. Plasmodium falciparum and Plasmodium vivax cause most of the malarial infections worldwide. Plasmodium vivax, Plasmodium ovale, Plasmodium malariae, and Plasmodium knowlesi are susceptible to chloroquine. Chloroquine was the world's most widely used antim...

Clinical Pharmacology of the Antimalarial Quinine in Children

Quinine is the best studied drug for treating severe malaria in very young children. Quinine may be administered in pregnancy and, at therapeutic doses, malformations have not been reported. Some strains of quinine from Southeast Asia and South America have become resistant. Quinine is the treatment of choice for the drug-resistant severe Plasmodium falciparum. The antimalarial mechanism of qui...

Quantification of labile heme in live malaria parasites using a genetically encoded biosensor.

Heme is ubiquitous, yet relatively little is known about the maintenance of labile pools of this cofactor, which likely ensures its timely bioavailability for proper cellular function. Quantitative analysis of labile heme is of fundamental importance to understanding how nature preserves access to the diverse chemistry heme enables, while minimizing cellular damage caused by its redox activity....
